Free and Glycosylated Emodin in Caulis Polygoni multiflori

Abstract:

Content of free and glycosylated emodin in Caulis Polygoni multiflori was determined in this paper. HPLC was used with C18 column (250 mm x 4.6 mm, 5?m), methanol with 0.4% phosphoric acid solution (85.5?14.5, V/V)) as mobile phase and the detective wavelength was set at 254nm. The speed is 1.0 ml•min-1. Emodin and rhein in the samples were well separated. The content of emodin was linear in the range of 6.5-52 ?g ml-1?r=0.9994?. The average recoveries for determining free and glycosylated emo-din were 97.2-99.7% (R.S.D %, 0.99-1.28%) and 95.7-96.4% (R.S.D %, 0.82-1.39%) respectively. The content of free and glycosylated emodin was 0.43 and 0.19 mg g -1 respectively. The content of rhein was very low, free rhein was not detected while the estimated value of glycosylated rhein was 11.38 ?g g -1. Caulis Polygoni multiflori was usually doceted with water in clinical application, so emodin and glycosylated emodin or emodin and the total emodin as index are more scientific and comprehensive for the quality control of Caulis Polygoni multiflori.
